John's wort extract produced benefits comparable to those of the tricyclic antidepressant drug imipramine, but with far fewer side effects. The 209 participants in the trial were randomly divided into two groups: one group received treatment with 600 mg St. John's wort extract three times a day, and the other took 50 mg imipramine three times a day. The 1,800-mg daily dose of St. John's wort extract used in the study and the 150-mg dose of imipramine were both twice as high as doses used in earlier trials that compared the two substances.
John's wort extract at a total daily dose of 900 mg was as helpful as the sedating tricyclic antidepressant drug amitriptyline (25 mg three times daily) in relieving symptoms in 165 people with mild to moderate depression. According to results of the randomized controlled study, there was no statistically significant difference in effectiveness between the two study groups, although the amitriptyline group demonstrated a tendency toward a better response rate. However, there was a significant difference between the groups with regard to adverse effects.
John's wort was just as effective as the antidepressant drug amitriptyline in treating symptoms of depression—with far fewer side effects. The multicenter, randomized, double-blind study involved 130 depressed people who took either 1 capsule of the herbal formula three times daily or 1 (25 mg) capsule of amitriptyline three times daily over a 6-week period. After 4 days of treatment, the dosage for both therapies was increased to 2 capsules three times daily, for 9 participants in the herb group and 5 in the antidepressant group.
